To me, it sounds like your editor might be mangling the template. Take 
the template where you only changed one letter and compare it with the 
original. You can do this with GNU diff:
   diff default/en/opac-main.tmpl default/test/opac-main.tmpl

...or maybe you have some other tool you prefer.
